About dendPoint

DendPoint is a novel computational method and database for predicting pharmacokinetic properties of dendrimer nanoparticles.

It is a machine learning approach that relies on a novel manually curated, literature derived relational database linking dendrimer physicochemical properties and pharmacokinetic parameters, including half life, clearance, dose in liver and urine.

Submission page - Single Prediction

How to run a prediction

To run a prediction:

  • Click on "Predict" (1) to open the submission page.
  • Provide Construct Properties (2).
  • Provide Surface Functional Groups information (3).
  • You can submit your query for analysis by clicking on (4).
Categorical and numerical descriptors:

  • Surface charge ranges from highly anionic [---] to highly cationic [+++], and was collected based on discussion in the respective manuscriptss of the number and type of surface charge and surface PEG loading.

  • Structural flexibility of the dendrimers ranges from relatively rigid [0] to highly flexible [+++] and was collected based on discussion in the respective manuscripts.

  • Generation values within the database range from 1 to 7, however predictions are not limited to this interval.

  • Construct Molecular Weight values within the database range from 2 to 334 KDa, however predictions are not limited to this interval.

  • Typical ranges for different Functional Surface Groups can be inspected by browsing the database, however predictions are not limited to these intervals.

Results

  • The predicted pharmacokinetic properties will be displayed at the top panel (1).

  • User have the option to run another prediction (2) or to run a comparison with another dendrimer (3).

  • A depiction of the dendrimer structure, generation and surface functional groups is also displayed (4).

  • In addition, a curve fort the predicted Plasma Concentration over time is available (5).

For Liver Clearance and Urinary Excretion a threshold of 20% defines whether a dendrimer is assigned as:
- Uptaken by liver/Excreted in urine (≤20%) or
- Limited liver uptake/Limited urinary excretion (>20%).

How to run a prediction

To run a prediction:


  • After clicking on "Compare" to open the submission page, provide the Construct Properties for the first (1) and and second dendrimer (2).

  • Provide the information regarding Surface Functional Groups for the first (3) and and second dendrimer (4).

  • You are then ready to submit your query for analysis by clicking on (5).

  • The predicted pharmacokinetic properties for the both dendrimers will be displayed at the top-left panel (1).

  • A curve fort the predicted Plasma Concentration over time for both dendrimers will be available at the top-right panel(2).

  • A depiction of the fisrt (3) and second dendrimer structure (4), depicting generation and surface functional groups are also displayed.
